摘要
目的 快速诱导藏红花球茎愈伤组织 ,解决藏红花资源短缺问题。方法 系统地研究外植体来源、培养基、温度、光照和激素因素对藏红花球茎诱导愈伤组织的影响。结果 以出芽后芽周围 2cm左右的球茎为外植体 ,MS为培养基 ,2 .0mg·L-12 ,4 D和 0 .2 5mg·L-16 BA为激素 ,在 ( 2 1± 0 .3 )℃ ,黑暗环境下培养 ,可以在 14d以后获得 60 %的愈伤组织诱导率。
OBJECTIVE: To rapidly induce Crocus sativus callus taking corm as explant. METHODS: The effects of corm source, various medium, temperature, light, and plant hormones on the inducing Crocus sativus callus were systematically and extensively investgated. RESULTS: The Crocus sativus corm callus was rapidly reduced within 14 days at (21 ± 0.3)°C, in darkness by taking the corm of 2 cm diameter around shoot as explant, MS as medium, 2.0 mg·L-1 2,4-D 0.25 mg·L-1 6-BA as plant hormones and the inducing rate reached 60%. CONCLUSION: The explant and the plant hormones were the key factors for callus induction.
