摘要 目的 测定人血浆中氟尿嘧啶的浓度。方法 高效液相色谱法 ,测量氟尿嘧啶的波长λmax=2 5 4nm ,流动相为水 甲醇 冰醋酸 ( 96.95∶3 .0∶0 .0 5 ) ,pH =3 .5。 结果 回归方程为Y =11.82X + 0 .0 78(r =0 .9992 ) ,其线性范围为 0 .66~ 42 .0 0 μg·mL-1,最低检测浓度为 0 .15 49μg·mL-1(S/N =3 )。结论 该法简单、快速、结果准确 。 OBJECTIVE: To determine the concentrations of 5-fluorouracil(5-Fu)in human plasma by HPLC. METHODS: The detection wavelength was at λmax = 254 nm. The mobile phase was H2O-CH3OH-CH3COOH(99.9 %) (96.95:3.0:0.05),pH = 3.5. 40 μL of the internal standard 5-Bru(200 μg&middotmL-1) was added into 200 μL of the plasma sample. Then 400 μL of methanol was added and centrifuged for 5 min. 20 μL of the supematant wereinjected into the HPLC system. RESULTS: The linear range of 5-Fu was 0.66-42.00 μg&middotmL-1, Y = 11.82X+0.078(r = 0.999 2) and the recovery was 99.77% with RSD 2.0% (intra-day) and 4.0% (inter-day). CONCLUSION: This method appeared bo te simple and accurate. It is suitable for the determing of drug concentrations of 5-Fu in human plasma in clinical use.
