摘要
目的 :比较亚洲型和西方型髓磷脂少突胶质细胞糖蛋白 (MOG)基因的多态性与比较亚洲型和西方型多发性硬化 (MS)患者。方法 :将 85例MS患者分为亚洲型 (4 0例 )和西方型 (4 5例 ) ,用微卫星法测定其MOG等位基因的多态性 ,并与 87例健康者 (对照组 )相比。结果 :MOG11/ 12等位基因与各型MS无关 (P >0 .0 5 ) ;亚洲型MSMOG5 1/ 5 2的 2 18bp等位基因频率显著高于西方型 ,西方型MS患者 2 18bp等位基因的频率显著低于对照组(P均 <0 .0 0 1) ;西方型MSMOG5 1/ 5 2的 2 2 2bp等位基因频率显著高于亚洲型和对照组 (P <0 .0 0 1~ 0 .0 5 ) ;亚洲型、西方型MS患者MOG5 1/ 5 2的 2 2 6bp等位基因频率均显著高于对照组 (P <0 .0 0 1~ 0 .0 0 5 )。 结论 :亚洲型和西方型是MS不同的临床亚型 ;MOG5 1/ 5 2的 2 2 2bp等位基因与西方型MS相关 ,与亚洲型MS无关 ;MOG5 1/ 5 2的 2 2
Aim: To explore the association of myelin oligodendrocyte glycoprotein (MOG) gene polymorphisms with multiple sclerosis (MS).Methods: A total of 85 patients of MS were allocated into Asian type(40 cases) and Western type(45 cases) . The MOG allelic polymorphisms of MS patients were detected by fluorescence labelling of PCR products and automated DNA sequencer. A total of 87 healthy unrelated individuals. Results: Asian type MS showed a significantly higher sex ratio(female/male), higher age of onset, and higher expanded disability status scale scores than those of Western type MS (all P <0.001). MOG11/12 alleles were not associated with Asian or Western types of MS( P >0.05). The frequency of 218 bp allele of MOG51/52 in Asian type MS was significantly higher than that in Western type MS ( P < 0.001 ). The frequency of 218 bp allele of MOG51/52 in Western type MS was markedly lower than that of healthy control ( P <0.001). The frequency of 222 bp allele in bothWestern type MS was significantly higher than that in Asian type MS and that of healthy control ( P <0.001~0.05). The frequency of 226 bp allele of MOG51/52 in Asian type MS and Western type MS was increased significantly compared with that of healthy control( P <0.001~0.005).Conclusion: Asian and Western types are 2 distinct subtypes of MS; the 222 bp allele of MOG51/52 is associated with Western type MS but not Asian type MS; the 226 bp allele of MOG51/51 may be a susceptical gene to MS.
