摘要
从临床分离绿脓杆菌菌株中 ,筛选与CMCC(B) 10 117株抗原高度相关的菌株PA0 2 0 10 ,PA0 2 0 15 ,PA0 2 0 33,PA0 2 0 37和PA0 2 0 4 8进行抗原成分分析。在此基础上 ,选择了PA0 2 0 4 8进行家兔抗原性试验和小鼠保护性试验。家兔抗原性试验证明 ,肌肉注射的效价为 1∶32 0~ 1∶6 4 0 ;静脉注射产生抗体能力强于肌肉注射 ,效价为 1∶2 5 6 0~ 1∶5 12 0。从动物保护性试验看 ,免疫动物均能很好耐受 5个LD50 的本菌攻击 ,在 10个LD50 的本菌攻击时 ,免疫动物的存活率能达到 90 % ;而对照组动物没有一个存活。所有试验表明 ,PA0 2 0 4 8菌株是替代CMCC(B) 10
In view of the fact that the strain CMCC(B)-10117(serotype 18 of IATS)is weakly antigenic when used in our producion of human polyvalent anti-Ps.aeruginosa plasma,5 strains of clinical isolates with antigens closely related to this serotype were used for screening with the purpose to select a substitute strain in stead of the original one.These strains were identified with 5 serotypes,i.e.,2,5,16,18and 20 by the followig tests.In rabbit antigenicity test,quantitative agglutination titers were obtained from 1:320 to 1∶640 by I.M.route and from 1∶2560 to 1∶5120 by I.V.route.Protection test showed a 90% to 100% protection against 5-10 LD 50and at the same time none of the control mice survived.Cross absorption test revealed that the strain PA02048 had a wider coverage of antigens.All these data showed the strain PA02048 is an ideal substitute for the strain CMCC(B)-10117 but the further proof would be awaited on the results of human immunization.
