抑制核桃组培中的褐化现象初探 被引量:23

摘要 以薄皮核桃茎尖为试材 ,对其分化生长过程中的褐化现象进行研究。经多次试验初步表明 ,为抑制褐化及保证茎尖正常分化生长 ,先将茎尖接种于 1 /2MS +2 g活性碳的培养基中 ,置于 5℃冰箱中黑暗培养7天后取出 ,再转接于DKW +6-BA 1 .0mg/L +IAA 0 .0 1mg/L +5ml2 0 %硫代硫酸钠的培养基中 ,置于 2 5℃室内光照环境下培养 ,每隔 1 5~ 2 0天转换 1次培养基 ,能有效地抑制褐化 ,确保核桃茎尖在整个培养过程中正常分化生长。 Experiment was aimed at studying the problem of browning control during the shoot-tip culture of pellicular walnut.Results showed that the beat way of preventive methods for the browning of explanted tissue is that the explants of walnut were cultured in the dark on DKW medium added with 6-BA 1.0mg/L + IAA 0.01mg/L +20% Na 2S 2O 3 5ml at 25℃ for 15~20d after dark culture on 1/2MS medium added with activated carbon 2g at 5℃ for 7d, and then, the subculture plantlets were transplanted on the same medium at intervals of 15~20d.
