阻断B7/CD28结合对同种异体小鼠移植心脏存活的影响

摘要 目的 :研究CTLA4Ig阻断B7/CD2 8结合对同种异体小鼠移植心脏存活的影响。方法 :建立同种异体小鼠移植心脏模型 ,将 2 8只受体鼠随机分为 3组。A组为对照组 (n =8) ,B组为CTLA4Ig注射组 (n =10 ) ,C组为供体脾细胞输注 +CTLA4Ig注射组 (n =10 )。观察小鼠移植的心脏存活天数和心脏病理改变。结果 :与A组相比 ,B组和C组的心脏存活时间明显延长 (P <0 0 0 1) ,C组心脏存活时间较B组也明显延长 (P <0 0 0 1)。组织学改变 :A组可见大量淋巴细胞、单核细胞浸润 ,组织充血水肿 ,心肌变性、坏死 ,出血 ,病理分级为Ⅳ级。术后 7d ,B组和C组则改变较轻 ,但仍可见多处片灶状炎细胞浸润病灶和心肌变性 ,病理分级为Ⅰ~Ⅱ级。结论 :CTLA4Ig可明显延长小鼠移植心脏的存活 ,而供体脾细胞注射能使CTLA4Ig的作用增强。
