摘要 目的 探讨雷公藤内酯醇对血管平滑肌细胞 (VSMC)增殖的影响及作用机制。方法 体外培养大鼠胸主动脉平滑肌细胞 ,通过3 H -TdR掺入试验 ,观察雷公藤内酯醇对血清诱导的细胞增殖的影响 ;采用流式细胞技术 ,观察该药对细胞增殖周期的影响。结果 雷公藤内酯醇在 (0 .5~ 4 )× 10 -5g/L的浓度范围内 ,以一种剂量依赖性的方式抑制了血清诱导的VSMC增殖 ;雷公藤内酯醇可显著增加细胞周期中G0 /G1期细胞百分含量 ,减少S期细胞的百分含量 ,阻断细胞由G0 /G1期向DNA合成的S期转化 ,作用呈剂量依赖性。结论 雷公藤内酯醇可能通过影响细胞周期的转化而抑制VSMC的增殖。 Objective It is to investigate the effect and mechanism of triptolide on vascular smooth muscle cells (VSMC) proliferation. Methods Rat tboracaorta VSMC were cultured in vitro. The influence of triptolide on serum induced cell proliferation was observed with the incorporation experiment of [ 3H] TdR. The influence of triptolide on cell proliferation cycle was observed with flow cytometry. Results Triptolide [(0.5~4)×10 -5 g/L)] inhibited serum induced VSMC proliferation with a dose dependent manner. Triptolide can increase the percentage of the G 0/G 1 phase cells and decrease the percentage of the S phage cells in the cell cycle, block the cell cycle transformation from G 0/G 1 phase to S phase. The effect showed a dose dependent relationship. Conclusion Triptolide might inhibit VSMC proliferation through influencing the cell cycle transformation.
