川芎嗪对新生大鼠离体胰岛分泌胰岛素的影响

Effect of Ligustrazine on Isolated Insulinism in Newborn Rats

目的 :了解川芎嗪对胰岛分泌功能的影响。方法 :取出生 3～ 5d的SD大鼠 ,无菌摘出胰腺 ,用V型胶原酶消化、分离纯化胰岛并用 1 6 4 0完全培养液稀释 ,按每孔 2 0± 3个胰岛分组接种于 96孔细胞培养板 ,观察不同剂量及不同作用时间下对胰岛素基础分泌及高糖刺激分泌的影响。结果 :在体外培养情况下 ,川芎嗪对新生大鼠离体胰岛B细胞胰岛素的基础分泌有明显促进作用 ,但对葡萄糖刺激的胰岛素分泌无显著影响。结论

Objective: To understand the effect of lingustrazine on insulin secretion. Method: Get SD rats aged 3 to 5 days for aseptic extraction of pancreas. Digest with V collagenase to isolate islets of pancreas, which was diluted with 1640 complete culture fluid. This was inoculated in 96-vent cell culture board with 20±3 at each to observe its effect on basal or glucose stimulated insulin secretion. Result: Under the condition of culture in vitro, lingustrazine markedly improved basal insulin secretion of B cells, but did not have evident effect on insulin secretion stimulated by glucose. Conclusion: Lingustrazine can stimulated iusulin secretion without affecting the normal secretion of islet cells.
