摘要
①目的 探讨哮喘小鼠是否存在Th1 /Th2免疫反应失衡。②方法 将BALBC 6周龄小鼠 30只分为对照组、卵白蛋白 (OVA)组 ,每组各 1 5只。OVA组第 1、1 4、2 1天给予用 1 .5mgAl(OH) 3乳化的OVA 1 0 μg腹腔注射致敏 ,对照组则在相应的时间给予 1 .5mgAl(OH) 3腹腔注射 ,两组均于第 2 6、2 7天雾化吸入OVA进行激发 ,第 2 8天处死小鼠行支气管肺泡灌洗 (BAL)同时摘取脾脏 ,用ELISA法测定支气管肺泡灌洗液 (BALF)和脾脏单个核细胞培养上清液中白细胞介素 4(IL 4)、白细胞介素 5(IL 5)、干扰素γ(IFN γ)含量 ,并计数BALF细胞总数及分类。③结果 第 2 8天OVA组脾脏IL 4含量为 (52 .1 9± 8.96)ng/L ,对照组为 (1 3 .33± 1 1 .1 6)ng/L ,两组比较差异有显著意义 (t=3 .40 8,P <0 .0 5)。OVA组BALF中IL 4含量为 (98.89± 39.30 )ng/L ,对照组为 (35 .0 6±1 6 .84)ng/L ,两组比较差异有极显著性 (t=3 .674,P <0 .0 1 ) ;OVA组BALF中IL 5含量为 (91 .55± 7.54)ng/L ,对照组为 (35 .33± 0 .69)ng/L ,两组比较差异有显著性 (t=3 .50 1 ,P <0 .0 1 ) ;OVA组嗜酸性粒细胞数量为 (56 .0 3±7.2 3)× 1 0 6 /L ,对照组嗜酸性粒细胞数量为 (1 .0 4± 0 .2 3)× 1 0 6 /L ,两组比较差异有显著性 (t=3 .687,P <0 .
Objective\ To investigate whether Th1/Th2 immune response imbalance existed in murine model of allergic asthma. \ Methods\ Thirty mice, aged 6 weeks, were divided into two groups: OVA group ( n =15) and control group ( n =15). For OVA group, sensitization was established by intraperitoneal injection of 10 μg of OVA emulsified in 1.5 mg Al(OH) 3 on days 1, 14 and 21. For control group, 1.5 mg Al(OH) 3 was given on corresponding days. On days 26 and 27, the mice of the two groups received aerosolization inhalation of OVA for excitation. Broncho alveolar lavage fluid (BALF) and spleen mononuclear cells (MNSc) were collected on day 28. The levels of interferon γ (IFN γ), interlrukin 4 (IL 4) and interleukin 5 (IL 5) of BALF and MNCs supernatant were measured by ELISA. The eosinophils (Eos) of BALF were counted.\ Results\ IL 4 levels of MNCs supernatant in OVA group were (52.19±8.96)ng/ L, and which were (13.33±11.16)ng/ L in control group, the difference was significant ( t=3.408, P <0.05). IL 4 levels of BALF in OVA group were (98.89±39.30) ng/ L, while (35.06±16.84) ng/L for the control, showing significant difference between the two groups ( t= 3.674 ,P <0.01); IL 5 levels of BALF in OVA group were (91.55±7.54)ng/L compared with (35.32±0.69)ng/L for the control ( t= 3.501 ,P <0.01). The Eos counts in OVA group were (56.03±7.23)×10 6/L, which were markedly higher compared with the control, namely (1.04±0.23)×10 6/ L ( t=3.687,P <0.01). Conclusion\ There is Th1/Th2 immune response imbalance, which polarizes toward Th2 type response, strongly suggests of existence of hyperaction of Th2 type immune response in asthma murine model.
