摘要
rRNA基因 (rDNA)序列分析方法用于植物系统分类与进化研究有较高的可靠性。近年来中药品种的鉴定也越来越多地使用DNA指纹标记 (如RAPD、RFLP、AFLP、SSR、ISSR、SSLP等 )和DNA序列测定。通过详述DNA序列测定中核基因组的rRNA基因 (rDNA)方法和 2 0 0 0年以来在中药鉴定中的应用 ,认为高等植物rRNA基因上的 18SrDNA、ITS及 5SrDNA片段常具有变异位点 。
It is reliable to apply rRNA (rDNA) sequencing in the phylogenetic classification and evolution study of plants. In the recent years, DNA fingerprinting (such as RAPD, RFLP, AFLP, SSR, ISSR, SSLP) and DNA sequential determination have been more and more widely used in the origin identification of Chinese materia medica. After a detailed description of nuclear rRNA genes (rDNA) in DNA sequential analysis as well as its application in the identification of Chinese materia medica sine the year 2000, the author came to conclude that heteromorphosis locus often exists in the 18SrDNA, ITS and 5SrDNA segments of rRNA genes of higher plants. There will be a vast range of prospects for applying it in herb breed identification, separating genuine herbs from false ones, and discovering new herbal sources.
