摘要
一组具有MADS box结构域的转录因子在控制花器官的诱导与发育中起着重要作用.以水稻广陆矮4号(OryzasativaL.Guang Lu AiNo.4)幼穗总RNA为模板,根据MADS box保守区的序列设计简并性引物,利用3′ RACE的方法获得了2个新的水稻花发育相关MADS box基因,分别命名为FDRMADS3和FDRMADS4;并利用5′ RACE的方法获得了该2个基因完整的cDNA序列,包括完整的编码区,5′ UTR和3′ UTR.它们编码的蛋白质都具有典型的MADS box结构域和半保守的K区,其与水稻中其他家族成员的MADS box结构域同源性高达90%以上,这说明它们都是典型的MADS box基因.
A group of regulatory factors containing the MADSbox domain plays an important role in controlling floral organ formation and development. In this study, total RNA from young inflorescence of riceGuangLuAi No.4 as template and a conservative DNA sequence of MADSbox to design primers were used, 2 new rice floweringrelated MADSbox gene cDNAs, named as FDRMADS 3 and FDRMADS4 respectively, were isolated by the 3'RACE methodology; and their fulllength cDNAs were obtained, including the whole open reading frame, 5'UTR and 3'UTR, by the 5'RACE methodology. Proteins encoded by these 2 genes contain a typical MADSbox domain that shows more than 90% identity to that of the other rice MADSbox proteins and a semiconservative Kbox, which means that FDRMADS3 and FDRMADS4 are typical MADSbox genes.
