摘要 目的 :研究WT1基因在造血系统恶性肿瘤中的表达。方法 :采用逆转录酶链反应 (RT -PCR)方法 ,测定158例血液系统恶性肿瘤患者及30例正常对照者WT1基因的表达。结果 :急性白血病 (AL)WT1基因阳性表达率为45.3 %。慢性粒细胞白血病 (CML)慢性期及加速期无WT1基因表达 ,而急变期WT1基因表达阳性。骨髓增生异常综合征 (MDS)WT1的阳性表达率为22.4 %。淋巴瘤及恶性组织细胞病WT1的阳性表达率分别为39.1 %和41.7 %。而正常人无WT1基因的表达。结论 :WT1基因在造血系统恶性肿瘤患者中高表达。随着疾病的进展 。 Objective:To study the expression of wilms'tumor gene(WT1)in hematopioetic malignanˉcies.Methods:Using the method of RT_PCR,the expression of WT1mRNA was detected in158patients with various hematopioetic malignancies and30healthy subjects.Results:The positive expression rate of WT1gene was45.3%(29/64)in patients with acute leukemia(AL).The patients with chronic myeloid leukemia(CML)in chronic and acederative phases did not express WT1gene;but when the CML patients progressed to blastic crisis,all of them did.The positive expression rate of the WT1gene was22.4%(11/49)in patients with mycelodysplastic syndrome(MDS).In patients with lymphona or malignant histocytosis,the positive expression rates of the WT1gene were39.1%(9/23)and41.7%(5/12),respectively.There was no expression of the WT1gene in normal controls.Conclusion:Aberrant overexpression of the WT1gene may contribute to hematopioetic neoplasia.The expression of WT1gene progressively increases as patient's condition worsen.
