摘要
目的:从分子水平探讨补肾健脾中药十四味建中汤治疗再生障碍性贫血(再障)的作用机制。方法:建立再障小鼠模型,通过抑制差减杂交,分离十四味建中汤治疗组小鼠和模型对照组小鼠之间脾细胞的差异表达基因。结果:分离出10个经十四味建中汤治疗后表达上调的差异表达基因,3个经十四味建中汤治疗后表达下调的差异表达基因。十四味建中汤促进免疫球蛋白α链恒区、α珠蛋白、β^(maj)珠蛋白、层粘连蛋白受体、谷胱甘肽过氧化物酶、核糖体蛋白S6等基因表达,抑制鸟氨酸脱羧酶抗酶等基因表达。结论:十四味建中汤治疗再障小鼠有效,可促进再障小鼠造血和免疫器官脾脏等功能的恢复。中医补肾健脾疗法的机制可能与上述基因表达变化有关。
Objective: To study the mechanism of tonifying kidney and invigorating spleen recipe-Shisiweijianzhong decoction in treating aplastic anemia(AA) on moleculal level. Methods:The AA models were induced through suppressed subtractive hybridization, spleen cell differentially expressed genes were isolated in mice with shisiweijianzhong decoction and the model control group.
Results: Ten up-regulated and three down-regulated differentially expressed genes were isolated respectively, Shisiweijianzhong decoction could promote the expression of Ig α-chain C region, α-globin genc,β-globin major gene, laminin receptor, glutathione peroxidase gene,etc,and inhibit the expression of ornithine decarboxylase antizyme gene,etc. It can conduce to the improvement of the AA mice spleen hemopoietic and immune function,etc.
Conclusion: Shisiweijianzhong decoction could effectivly treat AA, The mechanism may be related with above these differential genc expression.
