摘要
目的 分析经紫外线照射处理后日本血吸虫尾蚴蛋白的差异性表达 ,并从中筛选出有效的保护性抗原组分。方法 收集尾蚴 ,制备抗原 ,进行SDS PAGE电泳后 ,转印至硝酸纤维素膜上 ,与照射尾蚴免疫血清和正常感染血清分别进行免疫印渍 (Westernblot)试验 ,比较两者反应条带的差异性 ,筛选出惟有免疫血清所能识别的特有的抗原组分 ,并测定其分子量。结果 在童虫抗原中发现一条能被免疫血清所识别的特异性抗原条带 ,并测得其分子量为 75kDa。结论 紫外线照射致弱尾蚴可诱导宿主产生特异性抗体 ,被该抗体所识别的抗原分子量为75kDa。
Objective To study the different protein expression from ultraviolet irradiated cercariae of Schistosoma japonicum and screen for the effective antigens with protection against S.japonicum infection.Methods Cercariae antigens were prepared and separated by SDS PAGE electrophoresis then transferred to nitrocellulose membrane. Western blot was performed with irradiated cercariae immunized sera and normal infected sera.The positive bands probed with different immune serum were compared.A specific antigen recognized by irradiated cercariae immunized sera was screened and its molecular weight was determined.Results A specific antigen in cercariae was recognized by irradiated cercariae immunized sera, with a molecular weight of 75kDa. Conclusion Ultraviolet irradiated cercariae could induce host to produce specific antibody, which recognized an antigen with the molecular weight of 75kDa.
