大黄素甲醚对豚鼠皮肤黑色素细胞NOS的影响 被引量:6

EFFECTS OF EMODIN MONOMETHYL ETHER ON NITRIC OXIDE SYNTHASE IN MELANOCYTES OF GUINEA PIGS'SKIN

摘要 目的 :研究大黄有效成分大黄素甲醚对豚鼠皮肤黑色素细胞内一氧化氮合酶 (nitricoxidesynthase ;NOS)表达的变化 ,阐明大黄对黑色素细胞的有效作用浓度和可能机制。方法 :将 2 1只雄性豚鼠随机分成对照组和5个实验组 ,用大黄素甲醚 5种浓度对局部皮肤真皮注射处理 ,48小时后取材 ,免疫组织化学方法 (SABC法 )显示NOS的表达 ,用光学显微镜和图象分析仪对结果进行统计分析。结果 :大黄素甲醚作用下 ,豚鼠表皮黑色素细胞NOS表达明显减少 ,光密度明显下降 (P <0 .0 5) ;不同浓度药物作用之间无显著差异 (P >0 .0 5) ,加注侧与未加注侧之间无显著差异。结论 :大黄素甲醚对黑色素细胞NOS的表达具有调节作用 ,提示大黄对黑色素细胞的调节是经NO信号介导途径 ,为大黄的临床应用提供实验依据。 Objective: To study the change of nitric oxide synthase (NOS) expression in melanocyte of guinea pig's skin under the effect of emodin monomethyl ether(EME) and to explain the effective consistence and mechanism of rhabarb action. Methods: 21 guinea pigs were divided into a control group and 5 treatment groups at random. The treatment groups were injected in dermis with 5 different consistence of EME. The expression of NOS in skin melanocyte of each group was studied with immunohistochemcial method. Results: The expression of Nos in melanocyte was significanfly decreased(P<0.05). The different consistence of EME has no significant difference (P>0.05).Conclusion: The results show that EME has effect on the expression of NOS in melanocytes. The study exhibits that the effects of rhubarb on melanocyte were transduced by the NO signal transduction path way which may provide experimental material for further clinical use of this drug.
