摘要
目的研究肌层浸润性膀胱癌转移潜能异质性,并探讨生物通路在肌层浸润性膀胱癌异质性中的关键性作用。方法同位素标记相对和绝对定量(i TRAQ)技术和二维液相色谱串联质谱(2D-LC–MS/MS)量化和鉴定纯化不同转移风险组膀胱癌差异表达蛋白质,生物信息学方法进一步分析鉴定显著差异表达蛋白质并与既往相关文献的结果进行比较。结果生物通路的改变与浸润性膀胱癌异质性的调节密切相关。代谢相关通路参与了肿瘤的恶性生物学行为。显著改变的代谢通路包括糖酵解/糖异生通路、戊糖磷酸化通路、丙酮酸代谢通路、谷胱甘肽代谢通路等。结论遗传信息处理KEGG通路在决定浸润性膀胱癌恶性表型中起到重要作用。蛋白质量化分析结合KEGG通路分析有利于揭示膀胱癌发病机制并指导膀胱癌分子治疗方法。
Objective To study the metastatic heterogeneity of muscle-invasive bladder cancer(MIBC) and to explore the crucial role of biological pathway change in heterogeneity of MIBC cells. Methods i TRAQ and2D-LC-MS / MS were used to quantify and identify the differently expressed proteins in different metastatic risk groups. Then,the significantly changed proteins were further analyzed by using Arraytrack software. The interested proteins were compared with the data from published literatures to discuss the exact functions. Results The change of biological pathways was intimately related with cancer heterogeneity. Meanwhile,metabolic pathways play important roles in malignant biological behavior. The mainly altered pathways include glycolysis / gluconeogenesis,pentose phosphate, pyruvate metabolism pathway, glutathione metabolism, citrate cycle, etc. Conclusion The KEGG pathways of genetic information processing category play a crucial role in determining the malignant phenotype of MIBC cells. The quantification analysis of proteins combining with the KEGG pathway analysis contributes to screening candidate biomarkers and guides the biological molecular therapy of MIBC.
