MAGE-1相关肽抗原致敏DC活化的淋巴细胞对HCC细胞的体外杀伤作用

摘要 目的观察MAGE-1九肽致敏树突状细胞(DC)活化的淋巴细胞对HCC细胞的体外杀伤作用,确定MAGE-1九肽负载DC作为HCC疫苗的可能性。方法用MAGE-1九肽负载DC并活化T淋巴细胞,以不同效靶细胞的比例对正常肝细胞、黑色素瘤细胞、HCC细胞进行杀伤实验,3H掺入法测定杀伤率。结果用MAGE-1九肽负载DC并活化T淋巴细胞对黑色素瘤细胞及HCC细胞均有明显杀伤作用,而对正常肝细胞无杀伤作用;无关抗原负载的DC所活化的淋巴细胞对三种细胞均无杀伤作用;无抗原负载的DC其活化淋巴细胞对靶细胞亦无杀伤作用。结论MAGE-1九肽致敏DC活化淋巴细胞对HCC细胞有明显杀伤作用,MAGE-1基因可作为免疫治疗HCC的攻击靶点,MAGE-1九肽与DC联合可成为免疫治疗HCC的疫苗。 Objective To observe the kill effect of activated lymphocyte by DC pulsed with MAGE-1 nonapeptide to HCC cell lines ex vivo, and to evaluate the p ossibility of DC pulsed with MAGE-1 nonapeptide as a vaccine for HCC immunother apy. Methods Used activated T lymphocyte by DC pulsed with MAGE-I nonapeptide t o kill HCC cell lines ex vivo, and estimate the kill rate with 3H-TDR compared with melanoma cell lines and normal hepatic cells. Results 1,there is significan t kill effect on the melanoma cell lines and HCC cell lines with T leukocyte act ivated by DC pulsed with MAGE-1 nonapeptide, otherwise, no any action to the no rmal hepatic cells. 2, No killing action to all three types of cells with CTL ac tivated by DC pulsed with irrelevant antigen. 3, There is also no killing effect on target cells with lymphocyte activated by DC not pulsed with any antigen. Co nclusions T leukocytes activated by DC pulsed with MAGE-1 nonapeptide could ind uce kill action to HCC cell lines. MAGE-1 gene could be as a target attacked fo r tumor immunotherapy. Combined the MAGE-1 peptide and DC could be as a tumor v accine for HCC immunotherapy.
