黄芩苷下调CyclinB1基因表达对SMMC-7721裸鼠移植瘤的抑制作用

The Effect of Baicalin on Cyclin B1 Expression and Serum Alpha Fetal Protein(AFP) in Nude Mice Xenograft Model

目的:本研究通过对肝癌细胞株SMC-7721的裸鼠移植瘤模型使用不同剂量的黄芩苷处理,观察裸鼠移植瘤组织中Cyclin B1基因表达及裸鼠血清AFP蛋白含量的变化,初步探讨黄芩苷对肝癌的抑制作用,为中药黄芩的抗肿瘤应用提供理论基础.方法:使用传代后的人肝癌SMMC-7721细胞悬液建立裸鼠肝癌移植瘤模型,使用随机数字法分组.使用不同剂量的黄芩苷进行腹腔注射,2周后处死裸鼠解剖瘤体,使用免疫组化法测定肝癌组织中CyclinB1基因的表达水平、酶联免疫法测定各组裸鼠血清中AFP(甲胎蛋白)含量.结果:各黄芩苷组裸鼠的瘤荷显著低于对照组,黄芩苷组裸鼠抑制瘤肝癌组织中CyclinB1基因与对照组均有显著差异,并呈剂量负相关性(P<0.01);黄芩苷组裸鼠血清AFP阳性例数显著小于对照组,具有统计学意义(P<0.01).结论:黄芩苷对肝癌裸鼠移植瘤肝癌组织中CyclinB1基因有明显抑制作用,且其抑制强度与黄芩苷计量呈正相关关系;黄芩苷对肝癌裸鼠移植瘤外周血中AFP含量有明显抑制作用,其具体机制值得进一步研究.

Objective:T his research aim to investigate the effect of Baicalin on Cyclin B1 expression and serum Alpha Fetal Protein(AFP)concentration level of a orthotopic xenograft model of SMMC-7721 cell transplanted into nude mice.Method:Subcultured SMMC-7721 cell line suspension was used to establish nude mice orthotopic xenograft model.Random number method is used to assign treatment group.Different dose to Baicalin intraperitoneal injection was give to assign treatment group.Different dose to Baicalin in-traperitoneal injection was give to treatment groups.All mice were sacrificed 2 weeks later and Cyclin B1 expression and serum AFP level were evaluated.Results:All treatment group showed significant does cor-related smaller tumor burden,less Cyclin B1 expression and lower serum AFP level.Conclusion:Baicalin exerts significant dose -related inhibitory effect of Cyclin B1 expression and serum AFP level in nude mice model.
