肠三叶因子在胃黏膜应激性损伤中的修复作用 被引量:1

摘要 目的:研究肠三叶因子(ITF,intestinal trefoil factor,为三叶肽家族一员)在水浸束缚应激(WRS)大鼠胃黏膜基因表达的变化,探讨其在应激胃黏膜损伤中的修复作用.方法:采用单次及单次和重复水浸束缚应激制作模型,动态监测胃黏膜血流量(GMBF),大体及光镜下观察黏膜损伤程度(UI)及组织学变化,逆转录-多聚酶链反应(RT-PGR)检测ITF基因表达变化,免疫组化染色进一步证实ITF表达.结果:单次应激造成胃黏膜广泛损伤,但损伤指数在2、4、8 h逐渐减小,至8 h降为64.9%,GMBF逐渐恢复,至8 h上升为正常的89.8%,ITF基因表达增强(0.022±0.001vs0.177±0.010 P<.01),免疫组化染色计分为(0.134±0.001 vs0.253±0.01,P<0.01);单次和重复应激后胃黏膜产生适应性,胃黏膜血流量上升,损伤逐渐减轻,4次应激后,损伤指数降低为单次应激的22.0%,胃黏膜血流量上升为正常的94.2%,并且胃腺区细胞增生,ITF基因表达增强(0.040±0.001 vs0.372±0.01,P<0 01),免疫组织化学染色计分为(0.134±0.001 vs0.354±0.07,P<0.01).结论:ITF不仅可能参与胃黏膜早期重建(上皮移行),还有可能参与胃黏膜慢修复反应(细胞增生).
