摘要
目的 :探讨细胞因子 IL- 2、IFN- γ、IL- 4、IL- 1 0对 HBVDNA基本核心 C基因启动子 (BCP)变异、慢乙肝患者的病情及乙肝病毒 (HBV) DNA复制的影响。方法 :采用 PCR微板核酸杂交结合 EL ISA技术检测 HBVDNA BCP变异 ;荧光定量 PCR技术测定 HBVDNA含量 ;双抗体夹心 EL ISA法检测 IL- 2、IFN- γ、IL- 4和 IL- 1 0水平。结果 :1 HBVDNA BCP变异病人和非变异病人的 IL- 2、IFN- γ、IL- 4和 IL- 1 0水平无差别 (均 P >0 .1 ) ;2慢性重症肝炎组的 IL- 2、IFN- γ和 IL- 1 0水平高于肝炎后肝硬化和慢乙肝病人 (P <0 .0 5 ) ;而 IL- 4水平在各组无差别 (P >0 .0 5 ) ;3患者 HBVDNA水平随着 IL- 2水平的增高而下降 ,呈高度负相关 (r=- 0 .84 0 2 ;P <0 .0 1 ) ,其余 3种细胞因子水平与 HBVDNA水平未见相关关系。结论 :1机体的免疫压力可能对HBVDNA BCP变异无明显影响。2 IL- 2、IFN- γ和 IL- 1 0水平可能影响慢乙肝病情发展。3机体的 IL- 2水平对
Objective:To probe into the influence IL 2,IFN γ,IL 4 and IL 10 on the state of an illness, replication of HBV and occurrence of HBVDNA BCP mutation in patients with chronic hepatitis B.Methods:Microplate hybridization—ELISA and Fluorescence Quantitative PCR (FQ PCR) technology were applied to detect HBVDNA BCP mutation and HBVDNA quantity respectively. The level of IL 2,IFN γ,IL 4 and IL 10 in sera was investigated by ELISA method.Result:① The level of IL 2,IFN γ,IL 4 and IL 10 was not significantly different in the HBVDNA BCP mutation and no mutation groups (P>0 1).② The results show that the level of IL 2,IFN γ and IL 10 in CHH group was significantly higher than those in CH and LC groups (P<0 05), but no significant difference was Observed in the level of IL 4 (P>0 05).③The quantity of HBVDNA in sera of patients with chronic hepatitis B was lower with the elevation of the IL 2 level, and showed significant relationship (r=-0 840 2, P<0 01).Conclusion:① The immune pressure could be hardly relatied to the occurrence of HBVDNA BCP mutation. ② The level of IL 2,IFN γ and IL 10 might affect the development of chronic hepatitis B. ③IL 2 could restrain the replication of hepatitis B virus.
