摘要
目的研究犬体内丝裂霉素C(MMC)及葡聚糖微球(MMC-DMS)药代动力学。方法 20只杂种狗随机分为栓塞组和对照组,每组10只。栓塞组:X线透视下MMC-DMS经导管注入肝动脉,静脉取血,5.4 mg/kg微球。对照组:MMC,0.5 mg/kg,其他同前。3P97软件处理药-时数据,HPLC测定血药浓度。结果肝静脉中MMC动力学过程为三室,外周静脉血为二室;外周静脉血:栓塞组:MMC初值C=(0.3347±0.2870)μg/ml;对照组:MMC初值C=(0.8653±0.2047)μg/ml,二者比较差异有统计学意义(P<0.05)。结论 MMC-DMS血药浓度比较平稳,与单纯给以MMC相比,可降低外周药物毒性。
Objective To study on the pharmacokinetics of MMC and MMC-DMS in beagle dog.Methods 20 beagle dogs are divided into embolism group and contrast group,10 in each group.Embolism group:MMC-DMS are injected into hepatic artery by catheter in the way of X-rays and draw blood from vein,dose 5.4 mg/kg microspheres.Contrast group:MMC,dose 0.5 mg/kg,others same with former group.Drug-time data are processed by 3 P97 software and blood drug concentrations are determined by HPLC.Results The MMC kinetic process conforms to three-compartment model and two-compartment model in peripheral vein.Peripheral vein:the MMC primary blood concentration is(0.3347±0.2870)μg/ml in embolism group and(0.8653±0.2047)μg/ml in contrast group,with significant difference.Conclusion The MMC-DMS blood concentration is steady relatively and reduce peripheral toxicity in comparision with administration single MMC.
