摘要
目的:研究新制抗癌方合剂对染毒AFB1的HBV转基因小鼠肝癌变的预防作用及对氧自由基代谢的影响。方法:运用透射电镜等形态学观察方法和生化检测法研究新制抗癌方合剂对染毒AFB1的HBV转基因小鼠肝癌变的预防作用和对氧自由基代谢的影响。结果:模型对照组小鼠肝脏结节发生率和结节数目显著高于中药预防组(均P<0.01),模型对照组小鼠肝小叶结构消失,被大量不典型增生灶及增生结节取代,出现了大量异形细胞,异形细胞形态基本接近癌细胞,中药预防组小鼠肝小叶结构存在,可见少量肝细胞不典型增生灶和增生结节,异形细胞显著减少。中药预防组小鼠肝组织SOD、GST酶活性高于模型对照组,而MDA含量低于模型对照组(P<0.05或P<0.01)。结论:新制抗癌方合剂具有阻断AFB1协同HBV致肝癌变作用,其机理与该方提高肝组织SOD和GST酶活性,降低MDA含量有关。
Objective:To research the effects of New Anti-tumor Formular (NAF) on protecting against synergistic hepatocar-cinogenesis of hepatitis B Virus(HBV) expression and aflatoxin Bi(AFBi) and free radicals metabolism in HBV large envelope trans-genie mice. Methods:The methods such as transmission electron microsopy(TEM), biochemical analysis and so on were used to research the effects of NAF on protecting against synergistic hepatocarcinogenesis of HBV expression and AFB1 and free radicals metabolism in HBV large envelope transgenic mice. Results :The percentage of nodule occurrence and the quantity of nodule in the liv-er of the mice in the control group were respectivly significantly higher than that in the NAF group, which were treated by NAF. The structure of hepatic lobules in the control group dispeared instead of plenty of dysplasia proliferation. A lot of heteromorphic cells in the control group could be seen, which morphologies were similar to cancer cells' morphologies. There were the structure of hepatic lobules in the NAF group., There were only a few dysplasia proliferation in the NAF group. The quantity of heteromorphic cells in the NAF group decreased significantly compared to the control group. The activities of SOD and GST in the liver of the mice in the NAF group were higher than those in the control group( P < 0.01 or P < 0.05). But MDA content in the liver of the mice in the NAF group was lower than that in the control group(P<0.01). Condusion :NAF could protect against synergistic hepatocarcinogenesis of HBV and (?) bad relevance to increasing the activities of SOD and GST, decreasing MDA content in the liver of the mice.
