摘要
目的 探讨 Ig E高亲和力受体 β链 (high- affinity Ig E receptorβ gene,Fcε RIβ)基因启动子 - 10 9位 C/ T和编码区 Glu2 37Gly基因多态性与湖北汉族人变应性哮喘易感性及血浆总 Ig E的关系。方法 采用聚合酶链反应 -限制性片段长度多态性技术检测 FcεRIβ基因启动子区 - 10 9位和编码区Glu2 37Gly两位点多态性 ,采用病例 -对照法研究了 2 16例变应性哮喘患者和 198名对照。结果 (1)湖北汉族人变应性哮喘患者 Fcε RIβ基因启动子区 - 10 9位 T/ T、T/ C和 C/ C基因型频率是 0 .4 0 3、0 .4 91和0 .10 6 ;与对照相比差异无显著性 (χ2 =0 .384 ,P>0 .0 5 ) ,但变应性哮喘组 T/ T基因型患者血浆总 Ig E对数值 (2 .5 39± 0 .832 5 )与 T/ C基因型的对数值 (2 .2 78± 1.0 89)和 C/ C基因型的对数值 (2 .32 3± 0 .785 2 )相比差异具有显著性。(2 )变应性哮喘患者 FcεRIβ基因 Glu2 37Gly位点 Glu/ Glu、Glu/ Gly和 Gly/ Gly基因型频率为 0 .5 79、0 .370和 0 .0 5 1,与正常对照相比差异具有显著性 (χ2 =13.6 2 ,P<0 .0 1) ,变应性哮喘患者Gly/ Gly基因型血浆总 Ig E对数值为 (2 .6 2 2± 0 .9374 ) ,与 Glu/ Glu和 Glu/ Gly相比差异具有显著性。结论 Fcε RIβ基因启动子区 - 10 9位 T/ T?
Objective: To determine whether 2 polymorphism sites of the high-affinity IgE receptor β(Fc Ε RI β) gene were associated with allergic asthma in Han nationality of Hubei province in China. Methods DNA and clinical data were obtained from allergic asthma patients and were compared with those from a group of healthy control subjects. The subjects were genotyped for the -109C/T and a coding variant Glu237Gly in Fc Ε RI β gene by polymerase chain reaction-restriction fragment length polymorphism. Results: (1) The genotype frequencies were 0. 403 for -109T/T, 0. 491 for -109T/C and 0. 106 for -109C/C in allergic asthma patients in a Chinese population. No significant difference in the distribution of -109C/T polymorphism was found between allergic asthma patients and healthy control subjects; however, a homozygosity for the -109T allele was associated with increased total plasma IgE levels in patients with allergic asthma (F=7. 523,P<0.01). (2) The frequency of Gly237Gly genotype was 0. 051 in patient group, and 0. 020 in control group. The allele frequencies of Gly237 in the patients and control were 0. 236 and 0. 136 respectively. There was a significant association between Gly/Gly genotype and allergic asthma. Among allergic asthma patients Gly237Gly was significantly associated with high IgE. Conclusion: These data suggested that the Gly237Gly genotype of the Fc Ε RI β gene conferred genetic susceptibility to allergic asthma in Chinese, which affected the total plasma IgE levels in the allergic asthma patients. And homozygosity for the -109T allele was associated with increased total plasma IgE.
