脂氧素A4对重症急性胰腺炎模型小鼠肾损伤的保护性作用 被引量:2

摘要 目的采用小鼠动物模型明确脂氧素A4(LXA4)对重症急性胰腺炎(SAP)相关肾损伤的影响。方法研究分三组,分别为假手术组、SAP组及LXA4治疗组,选择30只雄性ICR小鼠,严格按照随机分配原则,保证初始每组为10只小鼠。通过逆行胰胆管注射法建立SAP的模型,并在术后1 h,LXA4组小鼠则按照LXA4 0.1 mg/kg剂量,通过尾静脉注射,建模后24 h处死小鼠,采用ELISA法检测标本血清中TNF-α、IL-10水平,对比各组小鼠外周血BUN及Cr水平,对比各组小鼠肾脏病理学评分。结果LXA4治疗组肾组织病理评分、血清TNF-α均可见显著低于SAP组小鼠(P<0.05),而血清IL-10表达水平则显著高于SAP组小鼠(P<0.05)。结论 LXA4可有效保护急性胰腺炎小鼠肾功能及抑制系统性炎症反应。 Objective To observe the effect of lipoxin A4( LXA4) on renal injury associated with severe acute pancreatitis in mice.Methods The study group was consisted of a sham operation group,an SAP group,and a LXA4 treatment group.A total of 30 male ICR mice were selected,and 10 mice were initially included in each group in strict accordance with the principle of random distribution. The SAP model was established by retrograde cholangiopancreatic injection.At 1 h after surgery,mice in the LXA4 group were injected with LXA4 at the dose of 0.1 mg/kg through tail vein.The mice were sacrificed 24 hours after modeling,and the specimens were detected by ELISA. The levels of TNF-α and IL-10 in serum were compared. The levels of BUN and Cr in peripheral blood of each group were compared,and the pathological scores of kidney in each group were compared.Results The renal histopathological score and serum TNF-α in the LXA4 treatment group were significantly lower than those in the SAP group( P< 0.05),while the serum IL-10 expression level was significantly higher than that in the SAP group( P < 0. 05). Conclusions Our study confirmed that LXA4 could effectively protect renal function and inhibit systemic inflammatory response in mice with acute pancreatitis.
