摘要
目的观察利拉鲁肽对内毒素急性肺损伤(IR)大鼠MOP、MDA、TNF-α、IL-6表达的影响,探讨其治疗IR的作用机制。方法选用Wistar大鼠60只,随机分为6组:正常组、IR组(急性肺损伤组)、利拉鲁肽低浓度组、利拉鲁肽中浓度组、利拉鲁肽高浓度组和地塞米松组,比色法测定各组大鼠肺组织匀浆中的过氧化物酶(MPO)及血清中丙二醛(MDA)含量。ELISA法检测各组大鼠血中TNF-α和IL-6的浓度。结果与正常组比较,IR组(急性肺损伤组)、利拉鲁肽低浓度组、利拉鲁肽中浓度组、利拉鲁肽高浓度组和地塞米松组5组肺组织中MPO和血清中MDA、TNF-α、IL-6炎症因子表达升高(P<0.05)。与IR组比较,利拉鲁肽低浓度组、利拉鲁肽中浓度组、利拉鲁肽高浓度组和地塞米松组4组肺组织中MPO和血清中MDA、TNF-α、IL-6炎症因子表达降低(P<0.05)。结论利拉鲁肽能降低内毒素造成的肺损伤过程中TNF-α和IL-6炎症因子水平,降低炎症反应危害。
Objective To observe the effect of liraglutide on the expression of MOP,MDA,TNF-alpha and IL-6 in rats with endotoxin-induced acute lung injury(IR),and to explore the mechanism of liraglutide in the treatment of IR.Methods Sixty Wistar rats were randomly divided into six groups:normal group,IR group(acute lung injury group),low concentration of liraglutide group,medium concentration of liraglutide group,high concentration of liraglutide group and dexamethasone group.The content of myeloperoxidase(MPO)in lung homogenate and malondialdehyde(MDA)in serum were determined by colorimetry.The serum levels of TNF-alpha and IL-6 were measured by ELISA.Results Compared with the normal group,the expressions of MPO in lung tissue,and the expression of MDA,TNF-a and IL-6 in serum in IR group(acute lung injury group),low concentration of liraglutide group,middle concentration of liraglutide group,high concentration of liraglutide group and dexamethasone group increased(P<0.05).Compared with IR group,the expressions of MPO in lung tissue,and the expression of MDA,TNF-a and IL-6 in serum in IR group(acute lung injury group),low concentration of liraglutide group,middle concentration of liraglutide group,high concentration of liraglutide group and dexamethasone group increased(P<0.05).Conclusions Liraglutide could reduce the levels of TNF-alpha and IL-6 inflammatory factors in lung injury induced by endotoxin and reduce the harm of inflammatory reaction.
