免疫性肝纤维化大鼠结缔组织生长因子、转化生长因子β1基因相关表达及汉丹肝乐的影响 被引量:3

摘要  目的 研究结缔组织生长因子(connectivetissuegrowthfactor,CTGF)mRNA与转化生长因子β1(transform inggrowthfactorβ1,TGFβ1)mRNA在免疫性大鼠肝纤维化中的表达状况及汉丹肝乐对其的影响作用。方法 用猪血清腹腔内注射复制免疫损伤性肝纤维化模型,造模同时给予汉丹肝乐口服作为肝纤维化防治组;免疫攻击12周后处死动物。原位杂交方法检测肝内CTGFmRNA、TGFβ1mRNA表达。结果 12周后模型组大鼠形成典型的肝纤维化,肝内CTGFmRNA与TGFβ1mRNA表达均显著增强,同时二者阳性分布部位相近,表达程度呈正相关性;以汉丹肝乐防治的大鼠,肝纤维化程度明显减轻,CTGFmRNA和TGFβ1mRNA表达的阳性指数下降,且CTGFmRNA表达与组织病理上肝纤维化变化呈正相关性。结论 CTGF与TGFβ1基因的增高表达与肝纤维化形成有密切关系;汉丹肝乐能有效抑制CTGF与TGFβ1基因的表达。通过在转录环节上阻断肝纤维化相关的细胞内信号转导途径,可能是该药物作用的重要分子机制之一。 ObjectiveTo study the expression of CTGFmRNA and TGFβ1mRNA in immunologically induced liver fibrosis in rats and effect of Handanganle on them MethodsMale wistar rats were given an intraperitoneal injection of porcine serum twice a week for twelve weeks. From the starting injection, Handanganle was fed on rats for precaution The animals were killed after the last injection. CTGFmRNA and TGFβ1mRNA were detected in liver sections by in situ hybridization The semi-quantitative results of the gene expression were analysed in contrast with the stage of hepatic fibrosis ResultsTypical liver fibrosis model in rats was shaped after twelve weeks of injection The positive stain of CTGFmRNA and TGFβ1mRNA increased in animals without Handanganle treatment and was distributed in the areas where fibrosis occurred There was obvious correlation between the strength of expression of CTGF and TGFβ1 In rats receiving Handanganle, the scores of CTGFmRNA decreased as the hepatic fibrosis mildened, the level of TGFβ1mRNA also declined ConclusionThe expression increase of CTGFmRNA and TGFβ1mRNA is correlated closely with hepatic fibrosis. Handanganle can effectively prevent increases in the gene expression of CTGF and TGFβ1One of the mechanisms of the intervention may be due to its blocking the intracellular signalling pathways involved in liver fibrogenesis
