氨氯地平和贝那普利对牛主动脉内皮细胞内皮素-1 mRNA表达的影响 被引量:1

摘要  目的 本研究利用牛主动脉血管内皮细胞的实验模型 ,观察氨氯地平和贝那普利对内皮素 - 1(ET - 1)mRNA表达的影响 .方法 制备 5~ 10代牛主动脉血管内皮的细胞悬液 .取生长融合、成片的细胞随机分成 3组 .分别为空白对照组、氨氯地平 ( 10 -5mol/L)、贝那普利组 ( 10 -5mol/L) .CO2 培养箱中孵化 2小时 ,收获细胞提取总RNA .用RT -PCR法扩增ET - 1cDNA的特异片段 ,PCR产物上样于 6 %聚丙烯凝胶行电泳 ,剥胶按改良的Brandt法进行银染 ,进行辉度扫描 ,对ET - 1的量进行定量分析 .结果 经区带密度扫描分析 ,与空白对照相比较 ,氨氯地平组和贝那普利组信号强度减弱 ( 0 .4 6± 0 .11vs 0 .2 6± 0 .0 5和 0 .4 6± 0 .11vs0 .2 3± 0 .0 5 ;p<0 .0 0 1) .而在两个处理组间无差异 .结论 氨氯地平和贝那普利可抑制培养牛主动脉内皮细胞的ET - Objective Endothelin-1(ET-1)contributes to the remodeling of the vascular wall and the progression of arteriosclerosis by constricting vascular smooth muscle cells and promoting their proliferation. Recently studies showed that angiotensin-converting enzyme inhibitors(ACEI) and calcium antagonists have protective effects on cardiovascular system.The aim of this study was to investigate the effects of on mRNA expression of ET-1 by cultured endothelial cells. Methods We tested the effects of Amlodipine(10 -5 mol/L) and Benazepril (10 -5 mol/L) on mRNA expression of ET-1 in quiescent bovine aortic endothelial cells(BAEC) in culture.The mRNA expression of ET-1 was dertermined by RT-PCR. The gel were scanned with a laser densitometer to determine the relating optical densities of the bands. Results Compared with vehicle control, both amolodipine and benazepril significantly decreased ET-1 mRNA expression(0.46±0.11 vs 0.26±0.05;p<0.001 and 0.46±0.11 vs 0.23±0.05;p<0.001).The ET-1 mRNA expression were no significant difference between amlodipine and benazepril groups. Conclusions These results suggest that in addition to their antihypertensive effects, amlodipine and benazepril may exihibit cardiovasculoprotective effects.
