睫状神经营养因子对视网膜色素变性小鼠光感受器功能影响的ERG分析 被引量:3

摘要 目的 :探讨在小鼠玻璃体腔内注射睫状神经营养因子(ciliary neurotrophic factor,CNTF)对视网膜色素变性(retinal degeneration,rd)小鼠视网膜光感受器的主要影响。方法 :将日龄为10d的12只rd小鼠随机分为CNTF治疗组、PBS(磷酸缓冲液)注射组及空白对照组。为CNTF治疗组小鼠在眼玻璃体腔内注射CNTF,为PBS注射组小鼠在眼玻璃体腔内注射磷酸缓冲液PBS,对空白对照组小鼠不进行任何干预,在三组小鼠日龄14d、18d、22d、30d时分别为其进行视网膜电图(ERG)检查。结果 :本研究观察了三组小鼠出生后14d、18d、22d获得暗适应后发生最大混合反应(Max-ERG)、获得明适应后发生ERG(Cone-ERG)及30Hz闪烁光ERG(Flick-ERG)的a波(图1)、b波(图2)的振幅,结果发现,与CNTF治疗组小鼠相比,PBS注射组小鼠在进行视网膜电图检查时上述振幅均较低,其潜伏期较长。在CNTF治疗组小鼠出生后第30d为其进行视网膜电图检查仍能记录到a波和b波。结论 :在rd小鼠玻璃体腔内注射CNTF能够有效延缓其光感受器细胞的凋亡,使其光感受器细胞功能得到显著的提高。
