以噻托溴铵和异丙托溴铵为例谈药物与受体结合的动力学特征 被引量:1

摘要 目的 :用噻托溴铵和异丙托溴铵作为例子,探讨药物与受体结合的动力学特征。方法 :通过对比噻托溴铵和异丙托溴铵在治疗慢性阻塞性肺病过程中与受体结合的动力学参数,分析药物结合动力学对药物作用的影响。结果 :当M3作为药物的靶标时,异丙托溴铵的Kd值是噻托溴铵的20倍。这说明,噻托溴铵的热力学活性要明显高于异丙托溴铵,而且其有效剂量明显低于异丙托溴铵。此外,通过观察t1/2(DR)值可以看出,噻托溴铵药理作用的持续时间远远长于异丙托溴铵。结论 :药物与受体的结合动力学参数可作为临床上优化选择药物的重要依据。
