摘要
采用荧光定量分析和过量表达的方法研究水稻Bowman-Birk家族基因BBTI-13基因的功能.结果显示,BBTI-13在野生型日本晴水稻中具有组织特异性,在胚和花药中表达最低;BBTI-13基因的烟草亚细胞表达定位表明其为组成型表达;过表达株系的田间表型分析发现,过表达转基因植株与野生型相比株型矮化,粒型稍变大.并通过对水稻株形矮化相关基因的表达进一步分析表明,BBTI-13基因的过量表达会引起水稻相关矮化基因的上调和下调反馈,其中Ghd2、DTH7、Ghd7、THIS1和Ghd8表现上调,OsBZR1、OsSIN表现下调.
The function of BBTI-13 gene which belonging to Bowman-Birk family in rice was studied by fluorescence quantitative analysis and gene overexpression.The results showed that BBTI-13 was tissue-specific expression in the wild-type rice,with the lowest expression in embryos and anthers.The subcell localization of BBTI-13 in tobacco indicated that BBTI-13 was constitutively expressed.The phenotypic analysis of over-expressed lines in field showed that the overexpressed transgenic plants were shorter than the wild-type rice,and the grain size was slightly bigger.Further analysis showed that overexpression of BBTI-13 gene could cause up-regulation and down-regulation feedback for rice dwarfrelated genes,for example,Ghd2,DTH7,Ghd7,THIS1 and Ghd8 were up-regulated,while OsBZR1 and OsSIN were down-regulated.
