人成骨细胞的I型胶原合成及基因表达 被引量:2

摘要 目的 :观察分析人成骨细胞在骨化三醇作用下细胞I型胶原的分泌。方法 :应用酶法分离细胞 ,四环素、茜素红鉴定 ,在成骨细胞生长的早期测定放射性元素掺入后细胞胶原的合成 ,原位杂交、mRNA表达反映细胞基因水平胶原的合成。结果 :1 0 (-8)mol/L骨化三醇明显提高成骨细胞内胶原的生成 (4 .43± 0 .2 3 ) ,杂交提示细胞胶原的灰度值为 (2 87± 2 5 ) ,Ⅰ型胶原α2 链表达 (3 .2 6± 0 .3 5 )和对照组有显著性差异 (P <0 .0 5 )。结论 :1 0 (-8)mol/L的骨化三醇在成骨细胞生长的早期 ,使细胞I型胶原的分泌明显的增加。 Objective:The scretion of type Ⅰ collagen in human osteoblast was studied by the 1alpha,25(OH)2D_3.Methods:The osteoblast was isolated from trabecular of the femur neck and checked up with tetracycline and alizarin red.In the early stage of growing osteoblast,the sythesis of type of Ⅰ collagen were mersured with radiation element.The expression of genes were showed in situ hybridation and mRNA.Results:1alpha,25(OH)2D_3, at a concentration range of 10(-8)M,increases sythesis of collagen intracellularly(4.43±0.23).The gray values are changed in hybridation(287±25).There was a significant increasing (P<0.05) in the mRNA expression of type Ⅰα_2 collagen(3.258±0.345)versus in the control group.Couclusion:1alpha,25(OH)2D_3 (10(-8)M) increased the scretion of type Ⅰ collagen in the early of stage of osteoblast.
