摘要
目的 :探讨淀粉样β蛋白 (amyloidβprotein ,Aβ)的毒性作用机制。方法 :使用MTT检验法检测细胞的生存率 ;使用Hoechst染色检测细胞的死亡类型。结果 :Aβ能够诱导人类胶质母细胞瘤细胞系U2 5 1死亡 ,自由基清除剂褪黑激素和谷胱甘肽均能显著降低U2 5 1细胞的死亡率。结论 :自由基参与Aβ的神经毒性 ,该毒性可以被自由基清除剂所阻断 ,从而为临床治疗阿尔兹海默氏病提供了新的途径。
Objective:To study the possible mechanism of amyloid β protein (Aβ) toxicity.Methods:MTT assay and hoechst dye staining were used to check the mortality and cell death type of U251 induced by treatment of A β. Melatonin and glutathione were used to block A β cytotoxicity.Results:A β could induce U251 cells to death. Both melatonin and glutathione could significantly lower the mortality of U251.Conclusions:Free radicals were involved in the cytotoxicity of A β and it could be partially blocked by free radical scavengers. It indicates a potentially new way to treat Alzheimer's disease.
