解毒祛瘀滋阴法防治实验性SLE类固醇性血脂异常

The Experimental Study of Jieduquyuziyin Methods(解毒祛瘀滋阴法) on Preventing and Treating Corticosteroid Hyperlipemia in Systemic Lupus Erythematosus

目的 :制作 SLE类固醇性高脂血症动物模型 ,探讨中医解毒祛瘀滋阴法对其防治作用。方法 :在中等含脂量饲料喂养条件下 ,MRL/lpr( 1 3～ 1 4周龄 )小鼠予以强的松 (每天 6mg/kg)连续4周 ,制作 SLE类固醇性高脂血症动物模型 ,并设正常中脂组和普食组作对照。然后 ,分设模型组(予生理盐水 0 .5 g/kg)、中药治疗组 (解毒祛瘀滋阴煎剂 37.3g/kg)、西药治疗组 (阿司匹林 90 mg/kg)。 1个月后 ,检测总胆固醇 ( TC)、甘油三酯 ( TG)含量。并用蛋白电泳技术对比分析胆固醇脂蛋白亚组分比率。结果 :模型组 TC、TG含量明显高于正常中脂组或正常普食组 ( P<0 .0 5 ) ;正常中脂组与正常普食组间无明显差异 ( P>0 .0 5 ) ;中药组和西药组 TC、TG含量均明显低于模型组 ( P<0 .0 5 )。中、西药组 LDL、VLDL、LP- a均明显低于模型组 ( P<0 .0 5 ) ,而 HDL高于模型组 ( P<0 .0 5 )。结论 :中等脂量饮食下 ,对 MRL/lpr小鼠灌胃强的松能制作出 SLE类固醇性高脂血症动物模型。中医解毒祛瘀滋阴法治疗 SLE的同时 ,能有效防治类固醇性高脂血症。

Objective: To make the animal model of cortiscosteriod hyperlipidma in systemic lupus erythematosus, and study the prevention and treatment effect of Jieduquyuziyin methods on it.Methods:The MRL/lpr mice (13～14 weeks old) were treated by prednisone(6mk/kg) for 4 weeks, being fed with middle doses of fat diet, in order to make the models of corticosteroid hyperlipidma in SLE. There were two teams of normal mice with middle or ordinary diet. Then, they were divided into 3 teams, model team being fed with 0.5ml physiological saline; TCM herbs team with Jieduquyuziyin decoction(37.3g/kg weight, each day);drugs team with Aspirin(90mk/kg weight). One month later, the levels of TC, TG, HDL-c, LDL-c and VDL-c were checked with auto-rapid electrophoresis.Results: The levels of TC and TG in model team were obviously higher than those in other two normal teams( P <0.05); The levels between in normal middle diet team and normal ordinary diet team were no obviously different( P >0.05); the levels in both the TCM team and medicine team were obviously lower than those in model team(p<0.05). The levels of LDL,VLDL,LP-a in TCM team and medicine team were obviously lower than those in normal team( P <0.05), while the levels of HDL in former teams were obviously higher than that in normal team( P <0.05). Conclusion:On the condition of middle fat diet, the SLE corticosteroid hyperlipidma animal model could be made by feeding MRL/lpr with prednisone. The cortiscosteriod hyperlipidma can be effectively prevented and cured when SLE was treated with Jieduquyuziyin methods of TCM.
