微阵列法检测上皮再形成过程中成纤维细胞基因的变化

摘要 目的研究真皮成纤维细胞在上皮覆盖过程中细胞因子基因水平的变化。 方法运用人cDNA微阵列技术 ,比较覆盖或不覆盖表皮层的真皮模拟物中成纤维细胞所含的细胞因子mRNA水平的变化。 结果成纤维细胞在 2 6 8个细胞因子基因中表达 6 8个基因 ,在 2 6 5个与细胞间相互作用相关的基因中有 6 4个被表达。第 7d时 ,4个基因上调 ,6个基因下调 ;第 15d时 ,4个基因上调 ,9个基因下调。 结论无表皮覆盖的真皮组织成纤维细胞中趋化因子的mRNA水平增高 ,金属蛋白酶mRNA水平下降 ,这有利于刺激上皮细胞的增殖和移动 。 Objective To learn the changes of cytokine gene levels in fibroblasts during re-epithelialization. Methods We used Atlas human cDNA expression arrays to study the level of cytokine mRNA in the fibroblasts of dermal equivalents with or without epidermis. Results 68 of 268 cytokines genes and 64 of 265 cell-interaction related genes were expressed in the fibroblasts. 4 genes up-regulated and 6 genes down-regulated on d7 whereas 4 up-regulated and 9 down-regulated on d15. Of all differential expressed genes, the most remarkable result was that chemokine genes up-regulated and metalloproteinase genes down-regulated on d15. Conclusion Fibroblasts in de-epidermalized dermis were inclined to secret more chemokine genes, which can stimulate epithelial cells proliferation and recruitment, and down-regulate metalloproteinase genes, and therefore might be useful for extracellular matrix remodeling.
