摘要 研究了用固相萃取预分离 ,高效液相色谱法测定杨梅树叶和树皮中杨梅素和杨梅素糖苷的方法。杨梅树叶和树皮中的杨梅素和杨梅素糖苷用 90 %甲醇加热回流提取 ,提取液用WatersSep Pak C1 8固相萃取小柱预分离脱脂 ,以WatersNova Pak C1 8( 3 .9× 15 0mm ,5 μm)色谱柱为固定相 ,0 .0 5mol/L磷酸二氢钾缓冲溶液和甲醇为流动相 ,在该色谱条件下 ,主要的杨梅素和杨梅素糖苷均达到基线分离 ;用紫外二极管矩阵检测器检测 ,并作了色谱峰纯度分辨。方法标准回收率为 95 %~ 10 2 % ,相对标准偏差为 1.5 %~ 2 .2 %。方法测定了几种杨梅树叶和树皮样品中的杨梅素和杨梅素糖苷 ,结果满意。 A high performance liquid chromatography (HPLC) method for the determination of flavonoids in Myrica rubra leaf and bark was studied. Flavonoids were extracted with 90% of methanol by heating circumfluence for 45 min, and then the extract was degreased by solid phase extraction with Sep-Pak-C18 cartridge. Flavonoids were separated on a Nova-Pak-C18 chromatogram column (3.9×150?mm), with methanol containing 0.05 mol/L potassium dihydrogen phosphate buffer solution as mobile phase at a flow-rate of 0.5 mL/min, and monitored with the photodiode array detector at 360 nm. Recovery of flavonoids was 95%~102%, with relative standard deviations 1.5%~2.2%. The method was applied to the determination of flavonoids in Myrica rubra leaf and bark with good results.
