摘要
目的 :观察微管稳定剂紫杉醇对肝癌QGY细胞的作用。方法 :检测紫杉醇对QGY细胞作用的时间效应和剂量效应 ,在光镜和电镜下观察紫杉醇作用后细胞形态变化 ;用流式细胞仪分析细胞周期的改变。结果 :随着作用时间及药物浓度的增加 ,紫杉醇对QGY细胞的增殖抑制作用就越明显。 5 6 μmol/L紫杉醇作用 72h ,细胞生长抑制率达 78.6 % ,形态学及流式细胞仪分析显示G2 /M期阻滞和细胞凋亡。结论 :紫杉醇诱发的人肝癌细胞的凋亡与细胞分裂期阻滞密切相关 ,这一作用机制将为临床治疗提供理论依据。
Objective:To investigate the effects of Taxol on QGY human hepatoma and its mechanism.Methods:The hepatoma cell growth was measured by MTT-microculture tetrazolium assay.Cell-cycle kinetics and apoptosis were analyzed by flow cytometry and microscopic examination.Results:Taxol inhibited the hepatoma cell growth in concentration- and time-dependent manners.Marked cells accumulated in G 2/M phase. The rate of cell growth inhibition was 78.6% after intervention of 56μmol/L Taxol for 72 hours. The cells showed G 2/M arrest and apoptosis.Conclusion:Taxol suppresses the growth of QGY human hepatoma cells in vitro by causing cell-cycle arrest, and apoptosis of the human hepatoma cells. The mechanisms of Taxol provide theoretical guidance for clinical treatments.
