摘要
经水抽提、乙醇沉淀及 DEAE—Sephadex A—50离子交换层析,从大麦芽中分离出酸性磷酸酯酶.酶比活力达1000u/mg.pr,经 SDS—PAGE 分析,该酶显示单一电泳染色带,分子量约为71kd,经聚丙烯酰胺凝胶等电聚焦测定,该酶等电点约为5.6.
An acid phosphatase with the specific activities 1000u/mg.pr frombarleys malt have been purified by extraction with water,alcohol precipitationand ion exchange chromatography with DEAE-Sephadex A-50.SDS-PAGE of the final preparation revealed a single protein staining bendwith a MW of 71kd.The pl value of 5.6 pH was determined by iso-electric focusing.
