HeLa细胞照射后存活后代生长特性及放射敏感性的变化

摘要 目的 :探讨HeLa细胞照射后存活后代生长特性及放射敏感性的变化。方法 :培养人HeLa细胞株和照射后存活的后代 ,测定其群体倍增时间、放射敏感性和细胞周期变化。结果 :HeLa细胞的群体倍增时间为 10 6 .0± 6 .8h ,HeLa细胞经 6MV X线DT 6Gy照射后存活后代群体倍增时间为 132 .8± 12 .2h(P <0 .0 5 )。照射前克隆形成率为 (12 .0±0 .5 ) % ,照射后克隆形成率为 (4.0± 0 .6 ) % (P <0 .0 5 )。照射前 2Gy生存分割指数 (SF2 )为 0 .5 2± 0 .0 5 ,照射后SF2为 0 .72± 0 .0 8(P <0 .0 5 )。HeLa细胞经照射后S期明显减少 ,G2 /M期阻滞 ,且呈剂量依赖性。结论 :HeLa细胞照射后存活后代生长延缓 ,放射敏感性增高。 Objective:To investigate the radiosensitivity and growth features of the progeny of irradiated HeLa cells.Methods:The HeLa cells were irradiated with 6MV X ray at dose of 0,2,4,6Gy and the progeny of the cells were cultured.The plating efficiency(PE),population doubling time(PDT),radio sensitivity index SF 2 and cell cycles of HeLa cells were detected pre and post irradiation.Results:The PDT,PE of HeLa cells before irradiation were 106.0±6.8h,(12.0±0.5)% respectively.The PDT,PE of irradiated HeLa cells at dose of 6Gy were 132.8±12.2h,(4.0±0.6)% respectively.The SF2 of HeLa cells was 0.52±0.5.The SF2 of irradiated HeLa cells was 0.72±0.08(P<0.05).HeLa cells showed significant dose dependent S phase decrease and G 2/M phase arrest after irradiation.Conclusion:Irradiation enhanced the radiosensitivity of HeLa cells.
