甘草酸抗人肺癌细胞增殖和侵袭作用的研究 被引量:7

摘要 目的 :探讨甘草酸对高转移人肺癌细胞 (PGCL3)增殖抑制和抗侵袭的作用。方法 :甘草酸处理PGCL3细胞 ,用细胞增殖抑制试验、软琼脂集落形成试验、侵袭、运动和粘附试验以及组织蛋白酶B活性测定 ,观察细胞增殖和侵袭能力的变化。结果 :甘草酸可降低PGCL3细胞增殖能力 ,并呈剂量依赖性 ,IC50 为 1.83mmol/L ;0 .5mmol/L和 1mmol/L甘草酸能抑制PGCL3细胞的侵袭能力 (P <0 .0 1) ,且有剂量依赖性。抗侵袭作用机理的分析结果显示上述浓度的药物对细胞的运动、粘附及组织蛋白酶B分泌均有明显的抑制作用(P <0 .0 1) ,软琼脂集落形成率也显著降低 (P <0 .0 1)。结论 :甘草酸有抗PGCL3人肺癌细胞增殖和侵袭作用 ,其抗侵袭机理不是对侵袭的某一环节的阻断 ,而是对侵袭各个基本环节都有抑制作用。 Objective:To study the effects on proliferation inhibition and anti-invasion of glycyrrhizin in highly metastasis lung cancer cell line (PGCL3).Methods:The proliferation inhibitive rate, the colony-formation rate in soft agar, the invasive ability to reconstituted basement membrane , the chemotafic migration ability, the laminin adhesion ability, and the activity of cathepsin B (CB) were tested to observe the effects of glycyrrhizin on PGCL3 cells.Results:Treated with glycyrrhizin, the proliferation of PGCL3 cells was inhibited obviously, and the inhibition degree was related to dose of the drug. IC 50 of the proliferation inhibition was 1.83mmol/L. Treated with 0.5 mmol/L and 1 mmol/L glycyrrhizin , the invasive ability, the adhesion and migrati on ability, the secretion of CB and the colony-formation rate in semi-solid agar of the PGCL3 cells were decreased significantly P <0.01), and the inhibition were in a dose-dependent manner. Conclusion:Glycyrrhizin can inhibit the proliferation and invasive ability of the PGCL3 human lung cancer cells. The mechanism of anti-invasion of the drug is to block many important points in invasive process, and does not limit any one point only.
