摘要
目的 :建立血清中蛇葡萄素的含量测定方法 ,并研究其在小鼠体内的药动学。方法 :血样经乙酸乙酯萃取后 ,采用高效液相色谱法 ,以乙腈 2 %醋酸 (1∶9)为流动相 ,在 2 90nm波长处检测。结果 :相关系数r =0 .9999,最低检测浓度为 56nmol·L- 1,日内及日间RSD分别为 1.0 2 %~ 3 .50 %和 3 .0 1%~ 5.54% ,萃取回收率为 91.97%~ 93 .3 2 % ,方法回收率为 99.41%~10 2 .10 %。药动学研究表明 ,蛇葡萄素的血药浓度 时间曲线符合一级吸收一级消除的单室模型。结论 :此法简便、可靠 ,灵敏度高 。
OBJECTIVE To establish a method for the determination of ampelopsin in mice serum and inve stigate the pharmacokinetic characteristics of ampelopsin in mice. METHODS Ampelopsin was extracted from serum with ethyl acetate and then injected to the HPLC system with a mobile phase consisting of acetonitrile-2% acetic acid(1∶9) and detected by UV detector at 290 nm. RESULTS The correlation coefficient was 0.9999 and the limit of detection was 56 nmo l·L -1 .The average relative deviations of within day and between day varia tion were 1.02% ~ 3.50% and 3.01% ~ 5.54% respectively. The averag e recovery of ampelopsin in samples was 91.97% ~ 93.32% .The recoveries of ampelopsin were 99.41% ~ 102.10% . The results showed that the serum co ncentration-time curve of ampelopsin was fitted to a one-compartment model wit h first order absorption and elimination. CONCLUSIONS The method was a simple,sensitive and accurate method for the determination of s erum ampelopsin levels in mice.It provides a suitable analytical method for its pharmacokinetic study.
