甲基强的松龙对特发性血小板减少性紫癜外周血淋巴细胞凋亡及FasFasl的影响

摘要 研究甲基强的松龙对小儿特发性血小板减少性紫癜 (ITP)外周血淋巴细胞凋亡及Fas(CD95 )、Fas配体 (Fasl)表达的影响及其临床意义。用流式细胞仪检测 2 5例ITP患儿治疗前后外周血淋巴细胞凋亡率及Fas、Fasl蛋白表达。结果 ,治疗前淋巴细胞凋亡率及Fas、Fasl蛋白表达与正常对照组相比差异无统计学意义 (P >0 0 5 )。治疗后血小板增加时淋巴细胞凋亡率增加 ,Fas、Fasl蛋白表达上调 ,与治疗前相比差异有显著性意义 (P <0 0 1)。结果表明 :甲基强的松龙可显著促进ITP患者外周血淋巴细胞凋亡的发生 ,上调淋巴细胞Fas、Fasl的表达 ,有助于清除激活的淋巴细胞。 To study the effect of methylprednisolone(MP) on the expression of Fas, Fas liga nd(FasL) and the apoptosis of peripheral lymphocytes and the role of them in the patients of idiopathic thrombocytopenic purpura(ITP). Flow cytometry was used to detect the expression of Fas and FasL protein and the apoptosis of lymphocytes in 25 ITP patients, before and after treatment. Results showed that apoptosis of peripheral lymphocytes and expression of Fas and FasL in the untreated patient were not significantly higher than those of normal controls( P >0 05). Apopt osis of peripheral lymphocytes, in ITP patients whose platelets increased after therapy, was significantly higher than that from normal controls and untreated g roup ( P <0 01). After therapy the expression Fas and FasL protein on lymphocytes are increased, as compared with that of untreated group( P <0 01). Conclusi on MP may increase the apoptosis of peripheral lymphocytes in treated ITP patien ts, the upregulating expression of Fas and FasL on lymphocytes after therapy may be help for eliminating the activated lymphocytes.
