摘要
对西瓜体细胞无性系变异进行了初步研究,结果表明:西瓜未成熟子叶和幼苗子叶的再生植株中存在明显的变异,其中以四倍体变异最为普遍,再生植株叶片气孔大小和保卫细胞内叶绿体数目可以作为从再生植株中早期鉴别四倍体植株的可靠指标。此外再生植株中还出现无权、浅裂刻叶和板叶变异类型,且有嵌合变异现象。RAPD分析结果表明,西瓜再生植株的基因组DNA在供试的部分引物上表现出DNA多态性。
Somaclonal variation of watermelon (Citrullus vulgaris Schrad) was studied. The results showed that apparent variation existed among the regenerants in vitro culture from the immature cotyledons, seedling cotyledons and euphylla of watermelon. In the variation plants, tetraploid was popular. The stomata size and chloroplast number in guard cells could be regarded as the reliable indicators of tetraploid in the early screening stage. Besides, the tetraploidy plants, other variations were observed, too. Some regenerated diploidy plants appeared morphologically abnormal on leaves and branches, some showed chimeric variation. RAPD analysis showed that polymorphisms existed among all regenerated plants.
