血浆中格列美脲的反相高效液相色谱法分析及其药代动力学研究 被引量:3

摘要 目的:建立血浆中格列美脲的反相高效液相色谱分析方法,并对其在人体内的药代动力学进行研究。方法:血浆中格列美脲用乙醚作液-液提取吹干后,再加入2,4-二硝基氟苯(DNFB)进行衍生化反应。反相高效液相色谱分析方法采用Shimadzu ODS色谱柱(4.6mm×150mm,5μm),柱温:30℃,流动相为乙腈-水(75:25,pH2.2),流速1.2mL·min^-1,检测波长350nm。结果:格列美脲在加-700ng·mL^-1血浆浓度范围内线性关系良好(r=0.9999),最低检测浓度为20ng·mL^-1。20,100,700ng·mL^-3种浓度的日内、日间RSD(n=5)分别为7.4%-10.5%和8.0%-11.8%;绝对回收率平均为75.0%,相对回收率平均为103.0%。结论:本实验建立的血浆中格列美脲反相高效液相色谱测定方法操作方便,结果准确,可用于该药进一步的临床研究。
