摘要
目的观察消风胶囊对小鼠迟发型超敏反应(DTH)的影响,并探讨其作用机制。方法采用2,4-二硝基氯苯(DNCB)背部皮下致敏、耳廓发敏的方法建立DTH小鼠动物模型,以双抗夹心ELISA法检测其血清中白介素2(IL-2)、γ干扰素(IFN-γ)、巨噬细胞培养液中肿瘤坏死因子α(TNF-α)水平。结果消风胶囊能明显抑制DNCB诱导的小鼠DTH(P<0.01),降低DTH小鼠血清中IL-2、IFN-γ水平(P<0.01,P<0.05),并能明显抑制腹腔巨噬细胞产生TNF-α(P<0.01),各剂量组间无明显的剂量相关性。结论消风胶囊有抑制小鼠DTH的作用,其机制可能与抑制了IL-2、IFN-γ、TNF-α等细胞因子的产生和(或)分泌有关。
Objective To observe the effect of Xiaofeng Capsule on the mouse DTH and explore its action mecha-nism on the mouse DTH.Methods The mouse DTH model was used and induced by the means of injecting subcuta-neously dinitrochlorobenzene(DNCB)and challenging DTH response with DNCB to study the effects of Xiaofeng Capsule on the levels of DTH and tested the level of several cytokines including interleukin-2(IL-2),interferon-gamma (IFN-γ)and tumor necrosis factor-α.Results Xiaofeng Capsule could inhibit DTH in mice induced by DNCB,decrease the lev-els of IL-2(P<0.01),IFN-γ(P<0.05)in serum of DTH mice,and obviously inhibit its peritoneal macrophage producing TNF-α(P<0.01).There is no obvious dose relativity.Conclusion Xiaofeng Capsule has the function of suppressing the mice DTH induced by DNCB.Its action mechanism may be related to the inhibitio n of the production and/or secretion of the cytokines such as IL-2,IFN-γ,TNF-α,etc.
