摘要
目的:克隆人肺癌转移相关基因片段。方法:采用mRNA差异显示技术对两株来源相同,转移能力不同的肺癌细胞株进行筛选和克隆与转移有关的基因片段,并采用通过GenBank对所获得基因片段,进行检索和序列分析及同源比较。结果:两株细胞的基因表达存在明显差异,共获得三个基因片段,其中片断1(P1)的序列与AND-34基因高度同源,片断2(P2)的序列与ARHA基因高度同源,片断3与目前已知基因均没有同源性,可能来自一个新基因的片段。结论:上述三个基因可能与人肺癌的转移有一定的关系。
Objective: To clone metastasis-associated gene tags in lung cancer cells. Methods: Metastasis-associated gene tags were isolated and cloned by mRNA differential display in two lung cancer cell strains from same origin but with different metastatic ability. Isolated gene fragments were blasted against database in the GenBank.Results:There were significant differences in gene expression between two cell strains. Three gene fragments were isolated. Of them, fragment 3 was a new sequence, fragment 1,3 were high homology with stromal cell protein (AND-34) and small GTP binding protein RhoA respectively.Conclusion:Isolated three genes may be relative to metastasis of lung cancer, and candidates of metastasis-associated genes.
