EB病毒活化端粒酶与p16^(INK4A)/E2F1调控的关系 被引量:2

摘要 目的 探讨EB病毒诱导人鼻咽上皮细胞永生化过程中 ,EB病毒活化端粒酶与EB病毒介导的细胞周期紊乱的关系。方法 利用WesternBlotting检测p16 INK4A和E2F1蛋白表达情况 ;利用hTERT -SEAP报道基因和端粒酶PCR -ELISA分别检测hTERT表达和端粒酶活性。结果 EB病毒下调p16 INK4A表达 ,并上调E2F1表达 ;与之相关 ,EB病毒诱导hTERT表达 ,并活化端粒酶活性。 Objective To study the mechanism of telomerase activation by Epstein-Barr virus in the immortalization of human nasopharyngeal epithelial cells.Methods The expression of p16 INK4A and E2F1 were detected with Western blotting analysis.The hTERT expression and telomerase activity were respectively tested with SEAP report gene assay and with PCR-ELISA analysis in human nasopharyngeal epithelial cells.Results The results showed that p16 INK4A expression was regulated down,at the same time,the expression of E2F1 was up-regulated by Epstein-Barr virus.And both of hTERT expression and telomerase activity increased in EBV-infected cells.Conclusion It is a preliminary study that EBV activated telomerase is through cell cycle to induce human nasopharyngeal epithelial cellular immortalization.
