硫酸镍对雌性大鼠卵巢的毒性作用及其机制 被引量:3

摘要 选健康性成熟Wistar雌性大鼠 ,每次以 5 0 0mg/kg、 2 5 0mg/kg、 1 2 5mg/kg硫酸镍 (NiSO4 )腹腔注射染毒 ,0 2ml/10 0g动物体重 ,1次 /d ,连续 2 1d。于最后一次染毒结束次日心脏采血放免法测雌二醇 (E2 )、孕酮 (P) ,同时测定卵巢NOS酶活力、NO含量及卵巢和子宫镍含量。结果染毒各剂量组动物子宫、卵巢镍含量及卵巢NOS酶活力与对照组比较差异有显著性 ;5 0 0mg/kg、 2 5 0mg/kg染毒组血清E2 、P含量 ,卵巢NO含量与对照组比较差异有显著性。提示腹腔NiSO4 染毒引起雌性大鼠卵巢损伤 ,血清E2 、P下降 ,其机制可能与卵巢镍含量升高诱导NOS。 Nickel sulfate was injected intra-peritoneally for Wistar female rats with sexual maturation daily at different doses(5.00mg/kg,2.50mg/kg,1.25mg/kg)for 21 days.On the 22 ndday,blood specimens were collected from the heart of the rats, nickel contents in the uterus and ovary,serum levels of progesterone(P)and estradiol(E 2)(with RIA),activity of nitric oxide synthase(NOS)and nitric oxide(NO)content in the ovary were measured.Results showed that the nickel contents in the uterus and ovary and activity of NOS in the ovary of the rats given nickel sulfate were higher than those in the control rats(P<0.01 or P<0.05);serum levels of E 2 and P in the rats with 5.00mg/kg and 2.50mg/kg of nickel sulfate were lower than those in the control rats(P<0.05 or P<0.01),and NO content in the ovary of the rats with 5.00 mg/kg and 2.50mg/kg of nickel sulfate was higher than that in the control rats(P<0.05).It indicates the ovary of the female rats was damaged by peritoneal injection of nickel sulfate,causing decrease in serum levels of E 2 and P,which was possibly associated with the increase in contents of NOS and NO.
