一氧化氮合酶在门脉高压大鼠肠黏膜中的表达及意义

EXPRESSION OF NITRIC OXIDE SYNTHASE AND THEIR SIGNIFICANCE IN ENTERIC AND COLONIC MUCOSA OF PORTAL HYPERTENSIVE RATS

目的 :探讨一氧化氮合酶 (nitricoxidesyethase ,NOS)在门脉高压大鼠肠黏膜中的表达及意义。方法 :将SD大鼠随机分成两组。实验组大鼠行门静脉两步结扎加左肾上腺静脉结扎 ,对照组为假手术组。门静脉完全结扎 2周后 ,应用免疫组织化学方法检测巨噬细胞型NOS(iNOS)和内皮细胞型NOS(ecNOS)在大鼠肠黏膜中的表达情况。结果 :门脉高压对大鼠肠道黏膜影响不完全一致。与对照组比较 ,实验组iNOS ,cNOS在小肠黏膜中高表达 ,iNOS在结肠黏膜中高表达 (P <0 .0 5 )。结论 :一氧化氮合酶与门脉高压大鼠肠黏膜局部病变有关。检测NOS对了解门脉高压性肠病有重要意义。

Objective:To explore the expression of nitric oxide synthase and their significance in enteric and colonic mucosa of portal hypertensive rats. Methods:Twenty male SD rats were divided into two groups randomly,and experimental group underwent two-stage ligation of portal vein. We detected the expression of iNOS and ecNOS in the enteric and colonic mucosa with immunohistochemical Strephavidin- Peroxidase method .Results:Two weeks after complete portal ligation, compared with the controls, the expression of iNOS and ecNOS in enteric mucosa were stronger, while iNOS in colonic mucosa was stronger.Conclusion:The changes of iNOS may play a role in the colonic mucosal lesion of portal hypertensive rats,while iNOS and ecNOS may play a role in enteric mucosal lesion.
