摘要
目的 探讨克隆法检测大鼠体细胞HPRT基因突变 ,为后续研究打下基础。方法 参照文献提供的克隆法 ,靶细胞为外周血淋巴细胞或脾淋巴细胞时 ,选取自体或异体的外周血淋巴细胞或脾淋巴细胞作为滋养细胞 ,比较相应的克隆率。结果 无论靶细胞为外周血淋巴细胞还是脾淋巴细胞 ,自体来源的滋养细胞组的克隆率高于异体来源的滋养细胞组 ,脾淋巴细胞为滋养细胞时克隆率较高。结论 自体来源的滋养细胞优于异体来源的滋养细胞 。
Objective To discuss the cloning method for detecting the HPRT gene mutation of rats. Method When peripheral blood lymphocytes or spleenic lymphocytes were used for target cells,and lymphocytes from homogeneity or heregeneity for feeder cells, cloning efficiencies were abtained. Results The cloning efficiency of the group with homogeneous lymphocytes for feeder cells was higher than that in the group with heregeneous lymphocytes.To select spleenic lymphocytes for feeder cells was better. Conclusion Homogeneous lymphocytes for feeder cells is better than heregeneous lymphocytes. Spleenic lymphocytes for feeder cells are suitable.
