p27和cyclinE在膀胱移行细胞癌中的表达及生物学意义 被引量:4

摘要 目的 探讨细胞周期依赖性激酶抑制蛋白 p2 7和细胞周期蛋白E(cyclinE)与膀胱癌生物学行为的关系。 方法 采用免疫组化SP法测定 6 5例膀胱移行细胞癌 ,5例不典型增生和 8例正常膀胱组织石蜡切片中 p2 7蛋白和cyclinE蛋白的表达。结果 p2 7在正常膀胱组织的阳性率为 1 0 0 % ,在不典型增生组织中阳性率为 6 0 % ,在膀胱癌组织中阳性率为 5 1 % (P <0 .0 1 ) ,p2 7在I、Ⅱ、Ⅲ级膀胱癌组织的阳性率分别为 75 %、5 4 %、2 6 % ,p2 7表达与膀胱癌的分级呈负相关 (P <0 .0 1 )。CyclinE在正常膀胱组织中的阳性率为 2 5 % ,在不典型增生组织中的阳性率为 6 0 % ,在膀胱癌组织中的阳性率为 5 1 %。CyclinE在I、Ⅱ、Ⅲ级膀胱癌组织的阳性率分别为 80 %、4 2 %、32 % ,CyclinE蛋白的表达与膀胱癌分级呈负相关 (P <0 .0 1 )。p2 7和CyclinE间在膀胱癌中的表达呈正相关。结论 p2 7和CyclinE与膀胱癌的组织学分级呈负相关 ,提示 p2 7和CyclinE可以作为判别膀胱癌分化程度的标志物 。 Objective To investigate the expression of p27 protein and cyclinE protein in human bladder carcinoma ,atypical hyperplasia and normal bladder tissue,and to explore the correlation of these proteins expression with biologic behavior of tumors.Methods p27 and cyclinE expression were studied in formalin fixed ,paraffin embedded sections of bladder carcinoma ,atypical hyperplasia and normal bladder tissue by means of a specific and sensitive immunohistochemical S P assay. Results The positive rates of p27 protein was 100% in normal bladder tissue,60% in atypical hyperalasia,51% in bladder carcinoma respectively( P <0.01),it was 75%,54% and 26% in Ⅰ?Ⅱ?Ⅲ grade of bladder carcinoma tissues respectively.The expresssion of p27 was negatively correlated with the histological tumor grades( P <0.01) .The positive rate of cyclinE was 25%,60%,51% in normal bladder tissue,atypical hyperplasia and bladder carcinoma respectively( P <0.01),it was 80%,42% and 32% in Ⅰ?Ⅱ?Ⅲ grade of bladder carcinoma tissues respectively.The expression of cyclinE was negatively correlated with the histological tumor grades( P <0.01).There was a positive relationship between the expression rates of p27 and cyclinE in bladder carcinoma. Conclusion P27 and cyclinE expression rates are negatively correlated with the malignant degree of bladder neoplasm, indicating that P27 and cyclinE might be an useful prognostic marker in bladder carcinoma.
